Echinoderms are abundant throughout the Indonesian marine water, especially in the coral reef ecosystem. Research on Echinoderms in Tikus island has been carried out since 2008. However, still, it has demanded to execute in-depth research in this region. The purpose of this research was to identify the diversity of Echinoderms in southern Tikus island where there is an ecosystem consisted of sand, coral reef, and seagrass. As the distance from the shore to the reef crest in Tikus island relatively wide, so it is quite easy for sampling process. Transect quadrant method was used implementing 1x1 m2 frame throughout the line from the shore to the reef crest with a total of 10 plots, and the distance between plots was 15 m. There was 5 point transect with a distance 50 m respectively. 127 individual of Echinoderms were sampled. The result revealed that 10 species of Echinoderms were found from 4 different classes, which are Archaster typicus, Culcita novaeguineae, and Linckia laevigata under the class of Asteroidea, Ophiactis savignyi, Ophiocoma erinaceus, and Amphiura sp. derived from Ophiuroidea class, Holothuria leucospilota under the class of Holothuroidea and Diadema setosum, Mespilia globulus and Laganum laganum derived from Echinoidea class.
