Abstract

The aim of the research was to know the capability of Bacillus licheniformis bacteria to produce α-amylase enzyme.This isolate which produced α-amilase enzyme was signed by clear zone around colonies after addition of iodine solution in medium containing 1% starch soluble. The activity of the α-amylase enzyme was measured by spectrophotometer at λ 540 nm and optimized at different pH, temperature, incubation period as well as 1mM solution of various monovalent and divalent cations. The results showed that the highest production of α-amilase activity was found at one-day incubation, which was 17.48 U/mL, while at 50°C and pH 8.5 the activity was 11.48 U/mL and 15.17 U/mL respectively. Experimental result of influencing metal ions showed that Ca2+ ion acts as an enhancer, while Na+, K+, Zn2+, Co2+ ions act as inhibitors of α-amylase enzyme activity.
