Abstract

Anggrek larat (Dendrobium phalaenopsis Fitzg.) is the identity flora of Moluccas Province, Indonesia. Based on Government Regulation no. 7 of 1999 on the preservation of plant and animal species, this orchid belongs to 12 species of protected orchids. In vitro propagation needs to be done as a conservation and fulfillment of market needs. Seeds are grown in Vacin and Went (VW) medium-enriched 1.0, 2.0, 3.0 g l-1 peptone. The characteristics of each of the developing phases in the germinated seed were observed and analyzed. For the establishing of proliferating cultures, intact protocorm of 2.7 months of age were growing on Murashige and Skoog (MS) medium with the addition of 4.54, 9.08, 13.62 mM Thidiazuron (TDZ) and 4.44, 13.32, 17.76, 26.64 mM 6-Benzylaminopurine (BAP), and control. The initial phase of development of seeds is divided into 6 phases that optimally grow on VW media addition of peptone 2.0 g l-1. Based on the value of the bud formation capacity (BFC) index, the optimal addition of TDZ 9.08 mM was used for shoot induction in protocorm of D. phalaenopsis with value 1.83±0.59.
